Treatment of cells with the macrolide antibiotic bafilomycin A1, an inhibitor of vacuolar V-ATPase, or with the lysosomotropic agent chloroquine, has been shown to pharmacologically inhibit autophagy as evidenced by an accumulation of autophagosomes, which in turn causes Bax-dependent apoptosis. However, bafilomycin A1 has also been reported to inhibit chloroquine-induced apoptosis. Chloroquine is a 4-aminoquinoline drug used in the treatment or prevention of malaria and as antinflammatory drug. Chloroquine at neutral pH is not protonated and enters the cell by simple diffusion, and diffuses to acid vacuoles too. These results indicated that PA also induced autophagy activation. Autophagy inhibition through chloroquine pretreatment or Atg5shRNA infection led to the increase of cell apoptosis after PA treatment. Moreover, induction of autophagy by pretreatment with rapamycin resulted in distinct decrease of PA-induced apoptosis.

  6. Chloroquine is an established antimalarial agent that has been recently tested in clinical trials for its anticancer activity. The favorable effect of chloroquine appears to be due to its ability to sensitize cancerous cells to chemotherapy, radiation therapy, and induce apoptosis. The present study investigated the interaction of zinc ions with chloroquine in a human ovarian cancer cell line.

In this paper, we have shown that pretreatment with chloroquine 0.3 mg/kg, i.v. bolus causes substantial reduction in biochemical and histological parametars and, also, in KIM-1 staining score. Chloroquine, well-known antimalarial drug, possesses some other, so called pleitropic effects antiinflammatory, anticoagulant and vascular actions. Oct 01, 2018 Chloroquine phosphate tablets should not be used in these conditions unless the benefit to the patient outweighs the potential risks. Usage in Pregnancy Usage of Chloroquine during pregnancy should be avoided except in the prophylaxis or treatment of malaria when the benefit outweighs the potential risk to the fetus. Chloroquine, a prototype anti-malaria drug, has been reported to possess anti-inflammatory effects. Moreover, chloroquine pretreatment could improve DNA damage repair. It is therefore reasonable to hypothesize that chloroquine pretreatment could attenuate ischemia/reperfusion injury in the brain.
